HPLC测定槐花药材不同部位芦丁和槲皮素含量

  摘要:目的 比较槐花不同部位(槐花、槐米及枝梗)中芦丁和槲皮素的含量。方法 采用高效液相色谱法进行测定,Ultimate XB-C18色谱柱(4.6 mm×250 mm,0.45 ?m),水-甲醇为流动相梯度洗脱,流速1 mL/min,柱温35 ℃,检测波长256 nm,进样量20 ?L。结果 芦丁含量为槐米>槐花>枝梗;槲皮素含量整体较低,且槐花>槐米>枝梗。结论 槐花各部位芦丁含量较高,且含量差别不大。槐花中的槲皮素含量明显高于槐米及其枝梗。

  Content Determination of Rutin and Quercetin in Different Parts of Sophora Japonica L.   Abstract:Objective To compare rutin and quercetin content of different parts (flower, bud and pedicel) of Sophora Japonica L.. Methods HPLC method was used with Ultimate XB-C18 column (4.6 mm×250 mm, 0.45 ?m), water and methanol as solvent system with gradient elution. The flow rate was 1 mL/min, column temperature was 35 ℃, detection wavelength was set at 256 nm, and injection volume was 20 ?L. Results The content of rutin was bud>flower>pedicel. The content of quercetin was low on the whole, and in sequence of flower>bud>pedicel. Conclusion In the different parts of Sophora Japonica L., rutin content is high with little difference. Quercetin content of flower is significantly higher than in bud and pedicel.

  槐花为豆科植物槐Sophora japonica L.的干燥花及花蕾,前者称“槐花”,后者称“槐米”,具有凉血止血、清肝泻火的功效[1]。现代研究表明,其黄酮类主要成分芦丁和槲皮素均具有多种药理作用[2]。近年来,随着医药工业的发展及保健品的开发,槐花作为黄酮类的提取原料,其需求量逐年递增。但因栽种局限性、生长半野生化及环保限制采收等原因使槐花产量减少[3]。市售槐花药材中除了槐花和槐米外,还含有大量的枝梗,在炮制过程中常常被作为非药用部位而除去[4]。本研究采用高效液相色谱法(HPLC)同时测定、比较分析同一来源的槐花药材中的槐花、槐米及其枝梗所含芦丁及槲皮素的含量,探讨槐花同一来源不同部位有效成分的差异性,为槐花药材的净制加

  Waters高效液相色谱仪(含1525工作泵及2487检测器);Adventurer-AR1140型电子分析天平。芦丁对照品(中国药品生物制品检定所,批号100080-200707),槲皮素对照品(中国药品生物制品检定所,批号100081-200907);槐花药材购于湖南长沙高桥药材大市场,经湖南中医药大学周日宝教授鉴定为豆科植物槐Sophora japonica L.的干燥花、花蕾及其枝梗;甲醇为色谱纯,其余试剂为分析纯;液相色谱用水为超纯水(自制,0.45 ?m滤膜滤过)。
