白及药材的HPLC指纹图谱建立及聚类分析

  摘 要 目的:建立白及药材的高效液相色谱(HPLC)指纹图谱,并对样品进行聚类。方法:采用HPLC法,色谱柱为Diamonsil C18,流动相为0.1%磷酸溶液-乙腈(梯度洗脱),流速为1.0 mL/min,检测波长为270 nm,柱温为30 ℃,进样量为10 μL。以1,4-二[4-(葡萄糖氧)苄基]-2-异丁基苹果酸酯为参照,绘制10批药材样品的HPLC图谱,采用《中药色谱指纹图谱相似度评价系统》(2004 A版)进行相似度评价,确定共有峰,并采用SPSS 23.0统计软件对其进行聚类分析。结果:10批药材样品的HPLC图谱有15个共有峰,其中9批相似度均大于0.90;经验证,10批药材样品HPLC图谱与对照指纹图谱具有较好的一致性。10批药材样品可聚为3类,S1、S2、S3、S4、S6、S8、S9、S10聚为第1类,S7聚为第2类,S5为第3类。结论:所建HPLC指纹图谱和聚类分析结果可为白及药材的质量控制提供参考。

  ABSTRACT OBJECTIVE: To establish HPLC fingerprint of Bletilla striata, and conduct cluster analysis. METHODS: HPLC method was adopted. The determination was performed on Diamonsil C18 column with mobile phase consisted of 0.1% phosphoric acid solution-acetonitrile (gradient elution) at the flow rate of 1.0 mL/min. The detection wavelength was set at 270 nm, and column temperature was 30 ℃. The sample size was 10 μL. Using 1, 4-two [4-(glucose oxy) benzyl]-2-isobutyl malate as reference, HPLC chromatograms of 10 batches of medicinal samples were determined. Similarity evaluation was conducted by using Similarity Evaluation System for Chromatographic Fingerprint of TCM (2004 A edition). Common peaks were determined, and cluster analysis was also conducted for it by using SPSS 23.0 statistic software. RESULTS: There were 15 common peaks in 10 batches of samples, and the similarities of 9 batches were more than 0.90. After validation, HPLC chromatograms of 10 batches of medicinal material were all in agreement with control fingerprint. The 10 batches of samples could be clustered into 3 categories. S1, S2, S3, S4, S6, S8, S9, S10 were clustered into category 1; S7 was clustered into category 2; S5 was clustered into category 3. CONCLUSIONS: Established fingerprint and the cluster analysis results can provide reference for the quality control of B. striata.

  白及為兰科植物白及[Bletilla striata (Thunb.) Reiehb. f.]的干燥块茎,其味苦、甘、涩,微寒;归肺、肝、胃经,具有收敛止血、消肿生肌之功效,临床多用于咯血、吐血、外伤出血、疮疡肿毒、皮肤皲裂等症的治疗[1]。现代药理学研究表明,白及具有止血[2]、抗菌[3]、抗肿瘤[4]、抗病毒[5]等多种药理作用。白及中含有多糖类、菲类、联菲类、联菲醚类、联苄类、甾体类和三萜类等化学成分[6],其中2-异丁基苹果酸葡萄糖氧基酯类的含量较高[7]。
  2015年版《中国药典》(一部)关于白及药材的质量控制标准中仅有理化鉴别、薄层色谱鉴别及水分、灰分检查等项目[1]。目前,对白及的研究主要集中在化学成分[6]、药理作用[2-5]、含量测定[7-8]等方面。但据笔者调研所知,目前白及的野生资源十分匮乏,药材主要源自栽培,而白及药材质量受生态环境的影响很大,并且市场上存在大量伪品(如黄花白及、小白及、华白及、竹叶兰、独蒜兰和舌苞兰等),使白及药材的有效性及安全性无法得到保障[9]。仅靠一两个化学成分的含量测定,并不能有效控制白及药材整体质量,而且该药材中许多成分的对照品在国家药品标准物质目录中均查询不到,这给同时检测其中多指标成分的含量带来了一定难度。指纹图谱是医药学界公认的控制中药材质量的有效方法,具有整体性和模糊性的特点。为此,笔者收集不同产地来源的白及药材,建立其高效液相色谱(HPLC)指纹图谱,并对得到的数据进行处理,结合聚类分析方法进行质量评价,为其质量控制提供科学依据。
